Look at the page source and search for rel="nofollow" at your link, also make sure the link is constructed using "<a href" and not with some Javascript code.
Download the website's robots.txt (xwww.otherdomain.com/robots.txt) or check it with a robots.txt editor on the web and see if the directory your link is placed in is not disallowed for the robots. (If your link can be found for example on the page: www.site.com/links/partners.html then look for entries in the robots.txt for the directory "links", and for the partners.html page too).

I love the SEO quake Toolbar. You can mark nofollow links which is very accurate and you can even find out if the page is cached or not in just a few clicks.

Edit: Just use the toolbar TrickyPlay mentioned, or the one from SEO Book.

If you do manual checking by reading the code, you should also look for
<META NAME="ROBOTS" CONTENT="NOFOLLOW">
It makes all links on a page nofollow.

Some people may also use meta tags for specific search engines to nofollow their links. Sometimes SEOs do this to "drop off the radar" of popular link-research tools, which mostly use Yahoo! for backlink research.

download this than right click the little q icon and select highlight no follow links. I leave mine on all the time but its quick and easy. Comes with a bunch of other features to but I like the nofollow one
